Oprettet i 1416 som kirke til Maribo Kloster, der blev nedlagt efter Reformationen i 1536. Kirken fik lov, at bestå og blev senere domkirke. Nordvest for kirken ses ruiner af klosteret. I Domkirkens krypt blev kong Christian 4.s yndlingsdatter, Leonora Christine (1621-1698) gravlagt.

Domkirken er oprindelig klosterkirke og i 1596 efter at byens daværende kirke var brændt blev den byens nye kirke. I 1804 blev den domkirke og i slutningen af det 19. århundrede blev tårnet bygget i forbindelse med restaurering af kirken.
